MLH1

MLH1 (MutL protein homolog 1) is expressed in normal proliferating cells and, when hetero-dimerized with PMS2, is involved in repairing DNA mutations which may occur during DNA replication. The antibody labels MLH1 protein in both normal and neoplastic cells. Lost expression or low levels of MLH1 are associated with colorectal and other cancers

Intended use

MLH1 is a useful tool for classifying tumors of the gastrointestinal tract, including associated extra colonic cancers such as endometrial and prostate cancers, when used with a panel of antibodies.

Assessment criteria
A moderate to strong nuclear staining reaction of the proliferating germinal centre B-cells in tonsil
At least weak to moderate distinct nuclear staining reaction of virtually all mantle zone B-cells and interfollicular lymphocytes in tonsil
In tissues with loss of MLH1, no nuclear staining reaction should be seen in the neoplastic cells, whereas a distinct nuclear staining reaction must be seen in stromal cells
