Human high molecular weight cytokeratins (CK HMW) is a term that collectively refers to the basic
cytokeratins 1 through cytokeratin 6 and the acidic cytokeratins cytokeratin 9 through cytokeratin 17.
Among those, CK HMW clone 34bE12 recognizes cytokeratin 1, 5, 10, and 14. These cytokeratins are
expressed in various epithelial basal cells and mesothelial cells.
Intended use
CK HMW clone 34bE12 is useful for differentiating benign prostate gland from prostate adenocarcinoma when used with a panel of antibodies. The mAb clone 34BE12 can not be used as general marker for CK-HMW due to cross-reaction with CK-LMW.

Assessment criteria
A moderate to strong granular cytoplasmic staining in all cell layers of the squamous epithelium of tonsil and esophagus
No staining reaction in hepatocytes and epithelial cells of the bile ducts in the liver**
*the most superficial squamous epithelial cells of tonsil and esophagus may show reduced staining reaction
**A moderate to strong and aberrant cytoplasmic staining reaction is seen in the epithelial cells of the bile ducts. The aberrant positive staining reaction most likely is caused by a cross-reaction of the mAb clone 34BE12 with a denatured form of CK19. The cells do not express CK-HMW |
